In this work the haemagglutinins of Periplaneta americana L. hemolymph have been characterized from the point of view of functionality, biochemistry, and localization. Three proteins, which are able to agglutinate rabbit erythrocytes, have been isolated and purified. They show different molecular weights (respectively 72000, 24000 and 22000 Daltons); one of them is certainly a lectin. Both cytoplasmic and membrane-bound localization of the three haemagglutinins has been pointed out by differential immunofluorescence in the hemocytes. Only the haemocytes, which show membrane-bound haemagglutinins, are responsible for cellular reactions (i.c.a.). The possible phylogenetical origin of the two groups of haemagglutinins is discussed.
