The localization of acetylcholinesterase (AChE) and pseudocholinesterase (PsChE) in adrenals of Rana esculenta and Bufo bufo was studied with Lewis and Shute method (1969). The activity of AChE in Rana is positive in the chromaffin cells and is localized in the nuclear envelope, in the cisternae of the rough endoplasmic reticulum and in some vesicles or cisternae of the vacuolar system, and in some chromaffin granules. The AChE reaction in Bufo chromaffin cells is less intense than in Rana's; it is positive in the vacuolar system; it rarely appears in the Golgi system and on the cell membrane for short lengths only. The nerve fibres located among the adrenal cells, both in Rana and Bufo, generally show a positive reaction to AChE located at axonal surfaces and in some vesicles and neurotubular structures within the axoplasm. The endings, whose morphologic aspect was cholinergic, generally react positively to AChE. No activity was found in Rana and Bufo adrenals when butyrylthiocholine was used as substrate.
