Cytological and radioautographic observations have been made on embryonic chick tibiotarsi cultivated in vitro in media containing, separately or together, H3-uridine ($5 \mu c/ml$) and actinomycin-D ($5 \mu g/ml$). It has been found that: a) condrocytes, even if not in division, actively synthesize RNA; b) RNA synthesis is completely inhibited by actinomycin-D; c) nucleolus is initially interested, and cytological lesions caused by this antibiotic are different from those observed with other antimitotics. These data have been considered in connection with the well known teratogenic action of actinomycin-D on skeletal development in the avian and mammalian embryos.
